Immune-modifying particles (IMPs) composed of biodegradable poly(lactic-co-glycolic acid) (PLGA-IMP) are taken up by circulating inflammatory monocytes via scavenger receptor MARCO and lead them to traffic the spleen where they undergo apoptosis. PLGA-IMPs has been demonstrated improve immune-mediated pathology in diverse tissues conditions. Here we report using intravenous for severe acute skin inflammation a C57/BL6 model injury induced topical nitrogen mustard (NM) exposure. significantly reduced edema delayed eschar formation NM-insulted mouse skin. show that daily administration PLGA-IMP post NM cutaneous insult not only decreased infiltration activation into wound area (p<0.025, n=5), but also resulted enrichment natural FoxP3+ regulatory T cells (nTreg) tissue as early day2 (p=0.022, n=5). To understand potential role nTreg on reduction cytokines eschar, well resolution inflammation, examined effects depletion/inhibition following anti-CD25 depletion. While reducing nTregs (p=0.0003, CD25 depletion expression immune markers such ST2, IL10-Tgfb, PD-1 (p<0.001, In addition, numbers IL10-Tgfb producing macrophages (p=0.0015, Infiltration wounds previously reported at days 5-7 during resolution. Importantly, our data shows treatment accelerates accumulation Tregs indicating it may serve novel disease modulatory treatment. We confident therapeutic protects mice from toxic exposure chemical insult.
